Effect of Mycoplasma hominis and Ureaplasma urealyticum on the Function of Astrocytes and Peritoneal Macrophages of Mice




Abstract
The purpose of this study were to invesitigate the use of culture method for detecting Mycoplasma hominis and Ureaplasma urealyticum in the various tissues of BALB/c mice which were intraperitoneally or intravenously per-inoculated with
Mycoplasma
hominis and Urealpasma uralyticum. Also, the effect of Mycoplasma hominis and Ureaplasma urealyticum antigens on the phagocytic activities and nitric acid production of the macrophages and astrocytes of mice were observed.
Mycoplasma hominis and Ureaplasma urealyticum were reisolated from the various tissues such as brain, liver, spleen, kidney, lung, lymph node, and blood from 6 through 24 hours after intraperitoneal or intravenous inoculation of Mycoplasma
hominis
and
Ureaplasma urealyticum in mice, but those organisms organisms couldn't confirmed from those tissues after 48 hours by culture method.
The phagocytic activities of the peritoneal macrophages and astrocytes stimulated with Mycoplasma hominis and Urcaplasma ureclyticum antigens were higher than those of the normal macrophages and astrocytes.
Nitric oxide production of peritoneal macrophages and astrocytes was increased by the combined treatlnent with IFN-r and LPS, or and Mycoplasma hominis antigen or IFN-r and Ureaplasma urealyticum antigen, whereas, nitric oxide production was not
increased by IFN-r, LPS, Mycoplasma hominis or Ureaplasma urealyticum antigen alone.
These results suggest that; I) Mycoplasma hominis and Ureaplasma urealyticum spreads to the vrious tissues of mice within 6 hours after intra-peritoneal or intravenous inoculation, ¥¢) the phagocytic activities of peritoneal macrophages and
astrocytes
were increased by stimulation with Mycoplasma hominis and Ureaplasma urealyticum antigens, ¥£) nitric oxide production of macrophages and astrocytes was augmented by the stimulated with IFN-r Mycoplasma hominis and Ureaplasma urealyticum
antigens.
Increased nitric oxide production of astrocytes is regarded to increase the intracellular bactericidal activites of astrocytes.
It is suggested, nonetheless, the inflammatory response of the Mycoplasm hominis and Ureaplasma urealyticum infected tissues is augmented by the increased nitric oxide.
